Increasing evidence suggested that side effects of the osteoporosis drug Prolia could increase the risk of hip fractures, femur fractures and other bone fractures, which typically occur following minimal or low trauma events.

MANUFACTURER: Amgen

OVERVIEW: Prolia (denosumab) was first introduced in 2010, as a treatment for osteoporosis. It was also the first drug every approved by the FDA specifically to treat postmenopausal osteoporosis. However, early on it was linked to an increased risk of bone fractures.

According to the consumer group Public Citizen, there were concerns over Prolia side effects dating back to at least 2009; a year before it was even approved to be sold.

In 2012, Health Canada and Amgen released a Prolia bone fracture warning letter, which was designed to alert Canadian users to an increased risk of atypical femur fractures that may occur with little or no trauma to the thigh bone; the thickest, largest, and purportedly strongest, bone in the human body.

Similar warnings were placed on Prolia sold in Europe. However, Amgen has decided U.S. consumers of Prolia do not warrant the same consideration or label warnings, despite the fact it is the same drug being sold in the U.S. as is being sold in Canada and Europe…just without those same warnings about the risks of bone fractures.

In 2013, the consumer watchdog Public Citizen added Prolia to its list of pills consumers should avoid, placing the drug on its “Do Not Use” list. Public Citizen noted that not only did Prolia triple the risk of hip fractures, femur fractures and other bone problems, but there were also concerns over infections and dermatologic adverse events.

In April 2019, Public Citizen petitioned the FDA to add a black box warning to Prolia over the risk of bone fractures. It is the most stringent label warning the agency can require a drug to carry.

When I was unable to tolerate the GERD from oral medications for osteoporosis, my primary care physician set me up with a clinic that gave me Prolia injections without ever telling me about the warnings. It wasn’t until I needed a tooth extracted that couldn’t be saved that I was given the horrendous warnings. I was then watched for dead bone disease and I fear future extractions as I have been unable to get off Prolia because I can’t afford the other injectables and insurance doesn’t cover. I have hip and back pain that make me miserable. I still haven’t been told there is a new medication I can tolerate covered by insurance. I hear it’s called the Prolia Trap. I live in fear of what is happening to my body and of the fact there is a time limit a person is able to take Prolia at all, in spite of the rapid bone loss when you stop.

None of this is news to the 9000+ members of the FAcebook Group, “Prolia (Denosumab) Side Effects and Support.” If there’s a cause of action, then why hasn’t someone started that action? There is clearly a class. There are clearly damages. There is arguably an inadequate failure to warn in the company’s materials to consumers and to physicians. The MSVFs and AFFs and MRONJ are consistently experienced, reported to physicians and to the FDA, and yet…no lawsuit. I’m a retired lawyer who was injured by Prolia, but I’m in Canada. In my jurisdiction, the manufacturer warnings were sufficient to interrupt the chain of liability, in 2018 when I suffered multiple spontaneous vertebral fractures from discontinuance without a relay. I had a cause of action against my physician, but not against the manufacturer. I suspect it’s the same in the USA. Scandalous.

Had my 2nd shot in April, I am so tired all the time, the pain my body has is horrendous. I had a bad back to begin with, now it’s horrible and can’t stand very long. I’m so sorry I agreed t this medication, even though my bone density is bad… I feel like I’ve been poisoned and I don’t know what to do. i also have so much joint pain, and my life is so diminished, it’s scary. Please don’t take this drug, it’s poison to our bodies.

I was prescribed Prolia injections in early 2022. I have had 2. So not let a GYN prescribe this without sending you to an endocrinologist first. My GYN was told that I have mild hypercalcemia. This injection has caused my blood calcium and urine calcium to skyrocket to where I am being advised to have parathyroid surgery. My risk of fracture has increased with Prolia. There are documented cases of Prolia causing Hyperparathyroidism . Beware.

I had my first Prolia injection in 2018 followed by 8 more. In March 2023 after many months of pain, I saw a maxillo facial surgeon and he confirmed that I was suffering of jaw necrosis. All implants will need to be removed in a few months and what will happen then, I have no idea. I simply can’t believe that a medication with so many terrible side effects can still be on the market and that doctors still recommend it to patients. Are they getting paid every time they prescribe it? Please stay away from Prolia and make sure you tell everyone about your experience.

I had three Prolia shots total. Then I stopped because I was suffering tooth loss due to bone loss in my jaw, left hip and left hip muscle detachment, overall pain in my entire body, strength loss, overall health loss. This is a drug would have eventually killed me. I feared my life… so I stopped. I am regaining strength in bone and muscle now. I would never take the drug again ever. 1500.00 a shot too.

Received January 11, 2023 painin Muscles bones bones and jaw. After 2 weeks. I was told if it would happen you would see in 2 or 2 days. NO one Everwood me about side effects. I call Prolis Pharmacy they told Mr it would be 2 weeks. I have written all this information At length i’m a form before this.

3 years ago I had my first injection of Prolia. Never again. I didn’t go back for another. I felt so ill, very tired, extreme pain in joints, especially in my jaw, it now feels like all the bones are broken in jaw and crack all the time. Doctor did not seem worried. I think the doctors need to research before giving to patients. I certainly wish I had read about it first. Do not take this drug, it is poison. There is a lawsuit going on, if you can afford a lawyer

I received the first Prolia injection on January 26, 2023. I had not researched this drug nor was I informed of the side effects and that I couldn’t just stop the drug without a relay drug. Within a few day of the injection, I had excruciating spinal pain, my knees began to swell, stomach muscle tremors. 2 months later and these side effects are still here. My quality of life has been diminished due to the pain of this drug. I will not take another injection and will fall into the “Prolia Trap” because I can’t take the other antiresorbitives due to current esophagus damage. Therefore, I will be at high risk of spontaneous spinal fractures. If I had done the research or had been informed by my doctor about this poison then I would have NEVER taken the injection. DO NOT TAKE PROLIA

I’ve had 2 Prolia shots six months apart. The first had some side effects, joint pain , lethargy, but I never thought about the shot causing it because the calamities started happening about a week after the shot. The second shot absolutely devastated me! I used the walk 5 miles, three to five times a week, hike the small mountains and parks around my area and swim in the lake. No more, the pain is horrible. The pain is in my hips, lower back, groin , thighs and knees. This drug company used me as an experiment and I am pissed! I am in physical therapy and trying to live my life as normal as I can, but I doubt I’ll ever be the same as before this poison was put into my body.

I took Prolia 7/19/22. Never knew I even got the shot. I got the second one on 1/23/23. Never ever again. The side effects are like a death sentence. Body aches and pain, joints popping, depression, dryness of tongue, insomnia, breathing, No strength whatsoever. It’s sad because I feel as if I’ve been injected with POISON!!

My problem with Prolia is not as severe as some of the comments posted here. My heart goes out to all of you. After receiving Prolia for several years I was never informed of the fact that once you start with it, even if your numbers are stable , you need to continue to get the injection again, later on. I was told that my numbers were better and I could stop it. Well last injection I believe was in 2017, my osteoporosis is worse than when I started the injections, much worse. Prior to the injections I was at risk for stress fractures, now what?

I took one injection and had muscle weakness and couldn’t hold my back up. Had a dental procedure that delayed the second shot. Once the second shot was given within two months the muscle weakness and bone pain started and palpitations high heart rate and then afib. Now the echo shows my left atrium mildly dialated and pulmonary pressure high now. I’ve been put on three meds and fear my heart will not be the same. Never had heart problems before the shot. Due for a shot in two weeks and I’ve now cancelled.

DO NOT TAKE PROLIA. My doctor wanted me to take the shots because of brittle bone syndrome and osteoporosis. To make a long story short I ended with a severe infection in my jaw my teeth chipped and broke off. Had to go into emergency surgery and had my right jaw removed now have titanium plate for a jaw. I talk funny and look weird, people can’t understand me.

I sm taking this med given the as certain that it was safe. i had My first in September 2022, my second is expected this March, My experience during this first injection has been with minimum noticeable side effects; I am concerned that the information from reviews always differ from that of the advice of the pharmaceuticals or the medicals offering the medications.

I was put on my first prolia injection at the end of 2019. I complained to my physician about headaches. He stated her never heard of this being a side effect of Prolia. My second injection was in May of 2020, 6 months later. In June I started with diarrhea. Couldn’t eat or sleep. Had severe abdominal pain, lost 10lbs in about 2 weeks. Had all kinds of urinary symptoms and GI symptoms. Went to the ER twice. Went on Ativan to sleep. Went into counseling because no one believed me that this was caused by the injection. So I was sick physically as well as mentally until I was do for another injection. The symptoms all went away by the time I was do for another injection. So far after reading other comments I’ve been OK except for my bone density. No One Should take this medication!

Hi there ,i took one injection on 4 /8/ 2022 , it has destroyed my body ,I have unreal body pains, my teeth are chipping ,my jaws are cracking, I can’t yawn , I went hoarse and a continuous tingling in my ear ,fatigue is unreal ,.I never knew that there was a black box warning either for this injection ,I have medical and dental records prior to this treatment .Take this injection off the market.

IMO, the real risk is Medication-related Osteonecrosis of the Jaw. The damage to my jaw is irreversible and there is a high likelihood that a surgical repair could cause further damage.

I have given this medication, Prollia, a chance since 2016 but I don’t think it’s working for me. My bone density continues to drop by -2. I’ve broken two ribs just getting out of bed. Do not take this drug. Do not take this drug unless the possibility of fracture is so high and the likelihood is great even tho I still cannot recommend. It is dangerous-rebound fractures, really? A drug that causes what you are treating? additionally it is a monoclonal antibody drug and there are ZERO peer reviewed studies ok-ing or even reviewing the safety of this drug in combination with COVID shots. Is it worth the risk until we do know? There are many new formulations of calcium that are better absorbed and easier to take made from sea life not the shells. Is it pricey yes- compared to less effective formulations-are you worth it yes! I had severe bone pain x4 months- have chronic pain In thoracic spine since injection (have NEVER had pain here before) tomorrow mri to see if I have rebound fracture/compression fracture (common with Prolia) I will NEVER take this horrible drug again. Try to warn other women of dangers: know one person who had to have her jaw replaced-can you imagine??? Luckily I only had one shot but I regret it every day. Please do your research. Your doctor does not know best, YOU do. Please. You do not want these side effects and even if you don’t know you have side effects, you could still have ongoing complications-additionally what if your insurance stops covering? What then -chances of rebound fracture especially thoracic spine compression fractures and hip fractures. Worth it? Finally before you ever get this shot, read up on the side effects and be sure to take calcium and vitamin d.

I had my second injection of prolia on September 28, 2022. I was bed ridden for 3 months with unbearable pain. I made trips to the E.R. for pain medication. I continue to have extreme pain and dizziness. Along with a host of other symptoms. I now am researching how to relay off of this horrible drug so as not to have the rebound fractures that accompany. It is scary. Since this drug is an injection, we as patients must rely on the doctor to alert us as to side effects but I fear that as in my case this is not done. I never saw a lable. I have lost all muscle tone at a time crucial to needing strength. I am still on extreme pain 4 months later and I am scared. Please help get this drug off the market. How could it help anyone.?

I was given Prolia in 2019 at 45 years old after breast cancer treatment. I began having hip pain and joint pain. It has been over three years and I continue to have the pain. It affects my life every day. As a mom, I can’t do things with my kids that I want to do. I have to take pain medicine if I am going to go anywhere. My quality of life has been affected. Since I can’t be active like I was, I have also gained weight. I can’t workout like I used to. No one knows what to do for the pain. I am tired of hurting!

Hi there, I had 3 prolia injections, experience muscle pain, hip knee pain, recently it was the worst , Hart time opening my yaw very painful, neck spasms couldn’t turn my next left or right, my legs are stiff and lots of spasms, I have ordered a wheel chair just in case, haven’t left the house for 3 weeks, couldn’t drive. Prolia is a very dangerous drug, I’m due for the next in February, NOT having another prolia

My Dr. Wanted me to take Prolia, my opinion they are not listening. I have not reached menapause,and won’t so long as I have adrenal mass. I already have bone pain, been allergic to the others, me and injections, medicines do not do well. I had enough anaphylaxis shocks, and injections are the worst. I have enough cardiac problems now, had neck and back fusions, I will refuse the appt. He made. I do not see passing the buck. To try and fabricate the problem. Advair, trelegy has caused the bone problem, good diet, calcium, magnesium, zinc, and D, added, and collagen, the natural. I do not need to gave more pain, to manage. My first dose of Prolia was March 2020. On October 23, 2022 my right thigh bone snapped while I was simply walking. I had to have a rod and 3 screws put In my right femur. I had no clue that thigh bone fractures was one of the uncommon side effects from Prolia until my sister who is a nurse told me after my surgery. After 27 months on Prolia I wish I never had taken it or at least known about the adverse side effects.

I took prolia injection I’m sick to stomach 24 7 and my jaw bones are falling out.I have headaches every day and night can’t sleep body hurts.I was not told of side effects And had my teeth pulled now im having reconstruction surgery to take dead bones out of my mouth.I look to have aged 15 yrs.Please help me I can’t take being sick every day Stomach pain is unbearable. I have had 2 injections of Prolia. The first one caused hip and leg pain but the second one not only caused the bone pain but I developed a severe throat infection with hoarseness. It has been 7 weeks since the last injection. I can’t wait until it is out of my system.

I was put on Prolia and took it for one Year. When Covid was at it”s worst I stopped getting the Prolia shots and went back on Fosamax. I’ve now had 2 spontaneous vertebral fractures since stopping Prolia two years ago. These are REBOUND FRACTURES caused by stopping Prolia. This has been well documented but I wasn’t informed of this risk before starting or after stopping the Prolia. I have been in pain for 10 months now and cannot stand or walk for more than about 10 minutes without severe pain.

I have been telling everybody I can to stay away from Prolia. In 2019, before I was due for my next injection (the third one) , I developed an abscess on my right jawbone. It was discovered then that I had necrosis of the jawbone and other mouth issues that required extensive surgery. As a result, I now have nothing to rest a bottom denture on. In the last couple of months, I have developed a growth on my lower left jawbone. I have made an appt. with my oral surgeons. Incidently, I stopped going to my first rheumatologist because after the surgery he wanted to put me back on Prolia. I refused and started seeing another doctor. If a class action lawsuit is initiated in the U.S., I would very much like to be included.

I have have two injections of prolia, I was told to not take any more it has ruined my teeth and gums , bones .within the past year I’ve paid over 7000. In dental . My balance is off I have to be careful not to fall . My knees , hips and joints hurt, feet hurt all the time . I’m 62 years old Im a floral designer I weigh 98 pounds . PROLIA IS A VERY DANGEROUS, HARMFUL DRUG. DO NOT TAKE IT!!! I had one injection of Prolia on April 11, 2022. Within hours bone started growing on top of my left foot, and a knot formed at my right distal radius wrist. When I get out of bed it is painful to walk. I fear that my left foot is permanently damaged. I live in fear that my left femur and hip may fracture from using Prolia. This drug should be banned from the market. I will not take another injection, nor will I take anymore medications prescribed for bone loss. My faith, nutrition, exercise, and will power are my only medicine. Drug companies are creating too many “designer, fashionable” drugs just to make money. Television ads promoting an annual injection to control cholesterol (sounds as dangerous as Prolia), a new drug for bone loss is being advertised. It’s just another Prolia with a different name. Amgen has severely harmed me.

After enduring 3 prolia injections,i choose to not have anymore..The first 2 where ok and after the 3rd injection ,2hrs later i was totally lethargic, as well my hair began to fall out/….I have worked in pharmacy for 30 years and i refuse to read side effects because each of us are different.. I will never choose to take this drug again,as my bones are still thining, major gum deterioation, as well as i am now in constant pain..

I see a general surgeon approximately every 6 weeks to excise painful lipomas primarily on my upper extremities from the use of Prolia. The numerous lipomas are painful, unsightly and replicate so that I can never get ahead of the re-appearance of them. A few incisions have become inflamed due to their location and suture material. I underwent a mammogram to determine that a lump in my breast was a lipoma not a cancerous tumor. I underwent a kyphoplasty on my lumbar spine due to a fracture there. My spine surgeon was very upset with me when I refused to continue this injectable and my rheumatologist gave me Fosamax. The spine surgeon will not operate on my spine until I have completed one year of Fosamax which is very upsetting. Alterations in the color and texture of the skin on my limbs make them susceptible to injury and bruises. I feel like I have lost 10 years of my life and become elderly overnight due to this drug.

I had a prolia injection in September 2022 I am in constant pain , my hips my legs I have no energy tired all the time . Had to give my job as a lollipop lady some days can hardly walk . Used to be so active for my 83 years I suffer constantly from urine problems burning when I wee very sore . This is my second injection and due for another one in March 2023 but since reading about Prolia I will skip that . My legs feel like dead weights .

After having two injections of Prolia the last being January 20th 2022, on February 17th my hip broke. I had to have screws put in my hip. When I woke up in the hospital my legs were swollen and have been ever since, the skin on my legs Weeps and I have water blisters on my legs. My hair is falling out I have migraine headaches my body is in constant pain and I can hardly walk. I have random shooting pains all over my body. I can’t really sleep at night more than a few minutes at a time. I’ve fallen multiple times because my balance is so bad and my legs are so swollen. No one at the doctor’s office told me anything about these side effects.

My 88 yr old mother has been on Alendronate for 4 yrs which did help increase her bone density by 12%, last bone scan showed only 2% reduction after being off 1yr now,Dr wanted to start on prolia,after reading the reviews from way too many people who have suffered with terrible side effects we made wise choice not to do at all,Alendronate stays in system for yrs after DC -ing .She hadn’t had any fxs prior to starting Alendronate & no side affects either luckily ,she said at her age with no fxs & Dx of osteopenia borderline osteoporosis would rather deal with that.We see it as this “with 10% still increased bone density after 4 yrs of taking Alendronate why change to something different if 1st TX plan worked,did research & OK to take what they call a “drug holiday” for few yrs from Alendronate then resume if needed in future.Prolia has only been used for 12 yrs compared to several decades of the bisphonates (Alendronate)& the bisphonates safer if have no side effects My advice is please do research before blindly agreeing to starting different Tx plan.Every patient has the right to refuse a TX option& should never be pressured by physican or scared into doing it,trust your initial gut feeling& do extensive research.I’ ve worked in medical field for several decades & aware of $ provided to physicians for using their products!! You can get 2nd opinion or change Drs,you get2 or3 estimates from companies before work done on home repairs,your body is more precious .There ARE good physicians out there who won’t pressure you to do something not comfortable with & take time to talk with you. Bottom line It’s YOUR decision & YOUR body not theirs.

My first and last prolia injection was February, 2021. It’s time for a follow-up with an endocrinologist. I was introduced to this drug via a referral. I was hesitant to receive a new injection but the endocrinologist assured me that she’d given “hundreds” of injections without any problems. I had an allergic reaction, drove to the local ER, had epinephrine and a relaxant, and was home later. Two weeks later I felt better. Now it’s August 2022, the drug has worn off, and I have severe hip pain and exhaustion. I already had lower back pain but the pain is souch worse. I’ve also had right hip screws from a previous fractured, and once again, the pain is worse. But for me, since I always deal with pain, the primary concern is unusual exhaustion.

I have just been prescribed Prolia and have yet to have my first injection. I have read some of the reviews and having this drug administered concerns me greatly. I am just healing up nicely after vertebrae compression fracture number six. I do have osteoporosis but feel very concerned at the side effects of this drug. What do I do ?

I’ve been on Prolia for many years after being diagnosed with osteoporosis. I have had many fractures since, I’ve got severe chronic pain that requires me to have service workers help me 7 days a week. I have lost many teeth and my teeth look disgusting. My life has been absolutely ruined by this drug that I thought was helping me. No one has ever told me about the adverse effects of this horrible drug. I stumbled on its faults only yesterday 9th August, 2022. I immediately went to a doctor to ask to stop taking this disgusting poison and I was sent for an immediate bone density scan which I won’t have the results for, for a couple of days. I’ve been told stopping the drug will cause even more bone’s to break and may stop me walking. What the hell!!!! Why is this drug even on the market. I’m soooooo upset!

I had my first Prolia injection about a month ago. A few days later I developed knee pains,muscle pains,back pain,spinal pain. The pain has ruined my life. I have to resort to using a cane to walk any distance at all. I have so much pain I can’t sleep. Before prolia I was doing 5km on a bike each day. Now I can’t even walk around my own house. I am at my wits end,I don’t know what to do. This prolia is poison.

I wish I had read about Prolia before starting. 2 1/2 years. I have lost 3 teeth, major cavities, my dentist asked me to talk to doctor regarding cavities, she couldn’t understand as I had never had cavities in the past. Bone loss around teeth. Also developed excema, excruciating leg and hip pain. Pre-cancerous cells, had to have laser treatment to remove. My health has been horrible since. I just made the connection by researching because next injection is due. NEVER use again.

I had my first Prolia injection in 2016 at the age of 51 after a Dexa scan showed lumber -2.2, left femoral-2.5. No fractures and a healthy 51year old, Rheumatologist said I needed Prolia due to the scans showing osteoporosis, I panicked and followed directions. I was not given any information regarding side effects or the fact that you needed a relay drug to come off Prolia. I had many side effects whilst taking Prolia, I had high cholesterol, stomach issues joint pain. I had countless blood tests, 24 hr heart ECG monitor twice, X-rays for pain, MRI’s to find cause of pain, sight irregularities (Eye specialist and MRI to find cause) stress test, sciatica, plantar fasciitis, trigger fingers on both hands, hip pain so bad I thought I needed a walking frame, could not bend down, could not stand unaided, turning in bed was a struggle, the list of things goes on and on. Prior to Prolia I did yoga daily but slowly I could do less and less if it until I could not get down on the floor to do it. It has wrecked my life and continue to struggle even after being off Prolia for 2 years. My health is improving slowly and hope it continues

I received prolia in April 12, 2022 after being pressured into it & with no warnings of the side effects. I have had horrific side effects & my body feels like it is in fire 24 hours a day. My health has declined & I now have a walker with seat prescribed to me & delivered as well as a handicap placard. I don’t know what else will happen in the future as a result of the prolia. I’m scared. I was healthy & only 46 when this happened. With a -2.8 t score & no spontaneous fractures. Only accident fractures in the past. I since have a new endocrinologist that said I should not have been put on prolia.

I had my first and only injection in April. I developed uncontrollable hives, eczema and rash that are debilitating. My left breast has grown out of proportion, I have headaches and lost my energy. I am 74 and prior to this shot I was active and full of life. Now I live in constant debilitating agony. The prescription meds make me tired and listless and I still itch. I can’t be outdoors or enjoy life any longer.

Prolia caused my teeth to rupture at the roots, my saliva ducts have stopped producing saliva and I have an ongoing painful tingling sensation in my jaw. I also have pain in my upper legs and hips and I am now bed bound at 53 years of age. I will never lead a normal life again. Prolia has ruined my life. Please contact me if there is a lawsuit that I can join. Thank you and best wishes to all of the other victims of Prolia.
