Increasing evidence suggested that side effects of the osteoporosis drug Prolia could increase the risk of hip fractures, femur fractures and other bone fractures, which typically occur following minimal or low trauma events.

MANUFACTURER: Amgen

OVERVIEW: Prolia (denosumab) was first introduced in 2010, as a treatment for osteoporosis. It was also the first drug every approved by the FDA specifically to treat postmenopausal osteoporosis. However, early on it was linked to an increased risk of bone fractures.

According to the consumer group Public Citizen, there were concerns over Prolia side effects dating back to at least 2009; a year before it was even approved to be sold.

In 2012, Health Canada and Amgen released a Prolia bone fracture warning letter, which was designed to alert Canadian users to an increased risk of atypical femur fractures that may occur with little or no trauma to the thigh bone; the thickest, largest, and purportedly strongest, bone in the human body.

Similar warnings were placed on Prolia sold in Europe. However, Amgen has decided U.S. consumers of Prolia do not warrant the same consideration or label warnings, despite the fact it is the same drug being sold in the U.S. as is being sold in Canada and Europe…just without those same warnings about the risks of bone fractures.

In 2013, the consumer watchdog Public Citizen added Prolia to its list of pills consumers should avoid, placing the drug on its “Do Not Use” list. Public Citizen noted that not only did Prolia triple the risk of hip fractures, femur fractures and other bone problems, but there were also concerns over infections and dermatologic adverse events.

In April 2019, Public Citizen petitioned the FDA to add a black box warning to Prolia over the risk of bone fractures. It is the most stringent label warning the agency can require a drug to carry.

After my first shot of Prolia, I started having bizarre problems with my jaw and couldn’t chew without biting my lip and tongue. I switched to soft foods and for the first time ever (at 50 years old), had to chew very slowly and I can no longer eat on public nor around anyone. I told my rheumatologist I did not want to do another shot but he dismissed the side effects which he never told me could happen in the first place. After the second shot, I lost my ability to walk and experienced profound issues with my jaw, teeth spontaneously coming loose, intense jaw pain, infection and gums spontaneously receding so much that the bone is exposed. My quality of life came to an abrupt end. I stopped Prolia 1.5 years ago, I am still experiencing major side effects from it and I am terrified of what’s next. I was never told of the profound side effects that could be caused by Prolia which I still experience every minute of every day.

I had my first prolia injection six months ago. I was NOT told of the risk of spinal fractures if you go off it. I had two compression fractures from falls 12 years ago, so now I am terrified that if I go off prolia I will suffer more spinal fractures. I want to go off it because I have had increased joint pain since I started on it, but now I feel trapped.

I was never told of any of the side effects or immune problems and I am immune comprised and have severe osteoporosis due to being on IV nutrition and having other health issues .. the doctor rx this knowing all this and withheld this information from me and knowing that I had previously fracture of back and neck.. having only one injection.. I am having severe pain in both hips and in my thighs and it’s beyond severe .. I cry.. very worried about fractures.. since my previous fracture still haven’t healed.. Then I have a severe comprised immune system due to being on Total parental nutrition.. very worried

I have had Prolia injections for a number of years with the next one due in 30 days. In Feb 2019 I incurred a T12 compression reaching for a product at the grocery. I had inj of Prolia that fall and another this past April. On 9/14/2020 I sat down hard to brake what would have been a nasty fall. Lost some skin on left arm. Started having low back pain that was relentless. This past Friday (10/23) I had kyphoplasty at L3. I had severe muscle spasms in back from inj in April. I plan on no more Prolia. But the horror stories of stopping this drug give pause to what my life will be hence. A sobering situation.

I received 6 shots of Prolia to prevent osteopenia, I was told there were no known side effects. After 6 shots my dentist became concerned about the changes in my mouth. I went to a referred specialist and he informed me that my jaw bones were affected by the Prolia and that I would never be able to have any kind of dental work that would require using the bones such as implants. I am now losing bridge work because the jaw bones are dying due to lack of blood flow.

I was on Prolia for four years, Just three months after stopping this medication I developed two compression fractures, I have been in a back brace for six months, the structure of my spine is permanently changed, The pain has been excruciating. This drug Needs to be taken off the market, no person Should ever go through this nightmare

I had 3 injections within an 18month period .. for a small spot of osteopenia on my hip. .I had severe bone pain while onnit so I stopped getting the injections .. A few months later I was in so much pain Went to emergency and had 2 fractures on my spine….Had 2 kypoplasty and 6 months later 1 more… I’m now taking Algae Cal.and Stronthium..Which has helped but still suffer wit back pain….Its Shaneful that this drug is still given in the US with no warnings… I hope that their isxa class action and this drug take off the market.

I have been on Prolia for 10 years I never noticed any side effects till I had dental implants it’s been 2years and have nothing but problems in my mouth sores bone pain in mouth it’s like an allergic reaction burning numbing tingling tongue has constantly welts been on steroids and 600 msg of gabapentin ..

I took my first…and last shot August 2020. In less than 3 weeks i started to have side affects, but i didnt put it all together till this week. When i went to see my oncologist, the one who prescribed this crap, she basically blew off my fears as stress, my age, etc… did set me up with a back xray, which showed osteoarthritis in my back. Anyway when my sister urged me to check out the Prolia, but util i was checking on drug interactions, i had never given it a second thought. I couldnt believe all that i found. All but 2 of my symptoms for the past 4+ wks were there. The shin pains that keep me up all night, the jaw pain where I cant even open wide enough to eat… the flu like feeling coming and going several days a week. There was numbness in my hands and dizzy spells at home and work. Some day i would call in because i couldnt even stand up with the pain in my feet and legs. There was stabbing pain in my ribs at anytime, for no apparent reason. This is crazy. This should be taken off the market.

Had first injection in March, 2020. I have suffered shoulder pain, back pain, left and right hip, both leg calfs, vision problems, numbness in both hands, and unable to get sleep. The Dr tried to convince me it is not the Prolia. No way. Tylenol or Aleve doesn’t give relief. I have called FDA and filed a report. Walking with a cane for balance. No more injections.

Had Prolia shot Oct,19,2020 and by Thanksgiving was having pain in both arms and hands and the17th of December,I left for California,just taking Advil.My right hand ,fingers were numb, then hand would tingle and sharp excruciating pain happened several times day and night keeping me awake.And backaches making it hard to get up and down .My left hand ached most of the time and was having charley horse cramps in both legs and feet.No pain medicine helped.When I returned home, went to my doctor , which sent me for exray of right shoulder, which came back as osteoporosis. Sent to have EMG done both arms and hands.Right hand came back as carpal tunnel and left hand as nerve damage .couldn’t wear watch or rings or wear a long sleeve that touched my wrist. Also went to Ortho Neuro for arms, had exrays, sent for therapy.After one session ,every thing shut down ,because of virus.Couldn’t have hand operated on,but he gave me a shot in the wrist, which helped plus Gabapentin.from family doctor.I’m still having problems after almost a year.Was never told of side effects,but knew the Prolia shot is the cause of all the pain I’ve had.I was a healthy woman of 83 years old ,on record as seeing my doctor for a sinus infection once a year. I will not ever take the shot again, and believe that a shot that was supposed to help, caused me the most pain I’ve ever had in my life.I would like to be informed if there is a lawsuit in the future.

I have had one shot……will be my last!! I had pain from before but this has put me to my knees!! All over I hurt..going up stairs I feel like I’m going to snap in half!! Itching in areas, extreme tiredness, some days I can’t even stand for more than 2-3 minutes!! My back, neck, shoulders, knees, ankles, arms, hands…….did I miss anything…. all ache!!! I don’t think all the doctors know what this really does to your body!! I’m in month 3 and can’t wait till this is out of my body!! Should have been more up front about the side effects……can’t see how FDA approved this…probably some greased palms in there!!! Can’t even trust them anymore…..not about health but money!!!

I have had one shot prolia I resisted it for a long time after reading horrible reviews I has a stress fracture in my foot Was advised by chemist and a prior doctor You only read the bad stuff online So I took a shot My left arm is extremely painful And fingers and thumb tingling all the time Pins and needles and one day extreme pain inside the bone of left arm Any advice please Good or bad I’m worried I am not going to take another shot I’m terrified

I started Prolia two years ago. I read about possible side effects, but was never told I could not go off this drug. The drug caused a substantial rise in my cholesterol, so I went on a vegan diet and was able to lower it somewhat. I do not want to continue taking this drug, will be telling my doctor I’m discontinuing it before my next injection. I do not want to take any other drugs for osteoporosis, and I feel that I was not given proper information about long term effects and not being able to stop taking it without risk of fracture. I feel duped.

When I completed Chemo and Radiation, my Doctor recommended Prolia to prevent fractures due to Letrazole medication. After 1 shot of Prolia, ( I refused any further injections) my bone and muscle pain have continually worsened. My jaw gets “stuck” and is painful. I did not have any of these issues before the Prolia. I actually felt decent for the first time since the end of Chemo then came the Prolia. I had a lung biopsy for a chronic cough (not sure if related) and was asked if I have Rheumatoid arthritis. The blood test showed Rheumatoid indications. I never had Rheumatoid indicators, not even with Chemo labs done every 3 weeks. I have taken steroids for seasonal allergies, heat rashes and then ibuprofen for this pain which helped. Now I’m having GI flare ups and was told not to take ibuprofen. I can hardly walk without pain in every joint, bone and muscle. My arms, hands, feet, shoulders, knees-all are effected. Maybe a broken bone would be less painful and would possibly heal. Not sure this will ever wear away.

Im female 53 – diagnosed with osteoporosis March 2019 and was given the prolia injection only once – pain in joints, hip, feet, fingers – everywhere basically! shooting pain in arms, anxiety, hair loss, vertigo 🙁 I didn’t go back for a second shot but a few months later thought I better do something to prevent further bone loss. Doctor gave me script for Risedronate, took 2 tablets over 2 weeks, I noticed my mouth area and lips tingling and feeling numb. I’ve not had another tablet since. 3 weeks later my mouth and cheeks lips are still numb and I’ve had an ear ache with ringing, shooting pain in arms and joint pain. The stress has caused anxiety and I’m on a down spiral, now I’m experiencing memory loss and I feel like I’ve aged about 20 years – Don’t know where to turn at this point. Tracy

I had my first injection on 1 May 2020. I am now suffering from vertigo. Extreme tiredness. I was not told of any side effects when given the injection. Never told my immune system could be compromised. My parathyroid reading went to 20 and my calcium is decreasing. I was never told this was chemotherapy. Suffering anxiety as to the future as I have CKD and coronavirus is escalating in my state.

I have been to several Othopedic Doctors and surgery is out of the question because I developed severe osteoporosis of the spine. L-2 down to S-1. Also, I have itching of the neck, ears, and scalp. Also bathroom issues. I’m afraid to stop because of the warning that the company said.

I had 7 rebound vertebral fractures after discontinuing Prolia. I wasn’t warned about this possibility by my doctor or Amgen. They occurred in 2017 through 22019. They were extremely painful and debilitating. I am permanently semi disabled now as a result of these fractures.

I stopped Prolia in February. I developed ringing in my ears and was afraid it was a side effect of Prolia. I was NEVER told that stopping Prolia could set set me up for vertebrae fractures, even though I specifically asked if there were any problem with stopping the shots. I developed a T6 fracture in May, suffered though the recovery and now have another one. The pain is extreme. I called the doctor’s office and told the employee who seems to manage the Prolia patients about my fracture and that my PCP showed me the information about stopping the shots causing these fractures. She had no idea this could happen and even said she needed to let people know who hadn’t gone in for their shots because of Covid. I was advised that by a different doctor that I needed to go back on the shots to try to prevent the fractures, I got another shot but am very afraid of more problems! When I started the shots I was told about possible jaw problem and femur fractures, but told the chances of problems were remote, I was NEVER told that once I started these shots I could not stop! This just shouldn’t have happened.

Jan’s Prolia story I have said EXACTLY the very same thing- I have all those issues but they are now at a level 3/4 and flare in sequences to a 7/8. 8 hours after my Prolia it all started – 12- 24 hours later I was in bed – crawling to the bathroom because my feet were so curled I could not walk … I had open sore rash on my face sizes of dime to Nickles- I was in bathroom 24-7 , vomiting, dizzy, huge mouth sores and a large fleshy mass in roof of my mouth. Severe headache and then the COPD cough- Coughing so much my abs and chest hurt… Dry skin and eyes – nails flakes off… hideous lady parts itching… eyes felt like cotton balls, and peeing and pooping myself- gastro issues , constant UTI , severe … lost much of my hair- bought a wig- hated wearing it… and my teeth issues- that was January 2015. I have said many times it ages me 10 years or more. And if it aged my outsides what did it do to my insides? Had to Quit my job 3 weeks later – I couldn’t manage to stop crawling and being in the bathroom until at least noon or later each day. Sobbed and cried by myself daily… – we got Coco a month later and she saved me. She got me out of bed, on afternoon walk and out socially… and kept me company. I owe her my life… Went to Mayo Clinic by myself with Mister for 9 days plus weekends … They addressed each single medical issue and stopped the treating my body as individual pieces to a whole body allergic and flared by Prolia. My doctor here followed their protocol to this day. I had rx for all of the above and My house is still full of all the rx. I can’t get rid of them just “in case” I need them. All due to one ridiculous PROLIa shot. Triggered every single autoimmune issue in my dna to attack my body all at once. I read on my Prolia sites people who are far worse … my hearts aches for them. And when I hear of someone’s family member I vow to share my story to not take that drug. I am a case study per Mayo Clinic for Fda and Amegen has my file as well. One day I hope to hold them accountable along with millions of other people.

After one shot of Prolia, I got psoriasis and severe pain in neck, shoulder, both legs, both hips. As a result I went to a chiropractor, an orthopedist, had an MRI for my shoulder, had physical therapy, took muscle relaxants and too much Advil, had acupuncture, went to the ER because I thought the back pain was maybe a kidney stone (it wasn’t) but the pain was so terrible they gave me intravenous Delaudid (a very heavy pain killer). Went to a dermatologist for the psoriasis on my face (now gone). And then to be told (I’m sure the drug maker purposely says this) that I can’t stop taking it ever or will have spinal fractures. Drug should be taken off market and doctors should be required to warn patients of these horrible side effects that are way too common with Prolia.

I had 2 shots of Prolia in 2015. After the first I felt weak and had aches and pains in my arms and legs. After the 2nd shot I started having terrible pains in my legs and hips, feeling very frail, balance problems from weakness, blood sugar problems and sinus problems. I had been very healthy and athletic all my life. Prolia took it away and made me a weak old woman. I can lay on my side for maybe 20 minutes before the pain in my hip wakes me up every night, roll over and in a few minutes the pain starts in the other hip and keeps me awake. Sometimes the pain in my thigh stops me from walking. The real kicker is that the doctor who gave me the shots said none of her patients ever had any problems, but she has collected thousands of dollars kickback from Amgen for Prolia. (look up Dollarsfordocs). This stuff needs to be banned!

I had one injection of Prolia October 7 2019. Within 3 hours muscle and joint pain began. This lasted 3 weeks. Next esophageal inflammation began. This is ongoing 6/2020. Gums inflamed. Severe hip and lumbar vertebrae pain. IGA dropped to 20. Remainder of immune system tests decimated. Prolia should be taken off market.

My mother took 3 injections of prolia which left her immobile. After she took the 2nd injection, she suffered from multiple vertebrae collapse and could not walk on her own. She had to use a walker for almost 6 months until the 3rd shot was given which totally made her bedridden. She cannot even stand on the ground with support. She is now on a wheelchair completely. This drug has given her so much pain. I am now trying to find a relay drug.

DO NOT TAKE PROLIA……I have taken it twice a year for many years through M.D. Anderson in Houston, TX. I trusted my oncologist even tho my bone scans came back significantly worse every time. I saw a world known Orthopedic Surgeons in New Orleans, Louisiana and he said NEVER take a Prolia injection again and he wished me luck with finding any doctor that would be willing to work on my teeth…..I believe I was nothing more than a human lab rat.

I have seen 6 dentists finally told I have ONJ last shot was November and I am in pain all day I have been given antibiotics. Rinses topical creams oxycodone and garbenpentin and nothing has helped with the pain I started out with one small area of bone exposed 2,months ago and now have another developing. I was first on forteo for a year. After that I was placed on Prolia and have been for a little over two years. I have always had stomach problems recently diagnosed with colitis. I had my front upper tooth loss at 13, and most all of my jaw teeth shortly after that over the years. I have lots of crowns and upper and lower partials. I have had a infection that was spread into the jaw bone and had to have the complete upper bone scrapped, guessing that was about 10 years ago. I’m 64 yr old now. My Dr. told me to go see my dentist before going on this drug and I did, no problem. I had no ideal!!!! Yes, my teeth were in the best condition that could be for me. BUT, if someone would have just ask simple questions, I should have never been on this drug!!!! Just based on my dental background. Now I’ve had 5 root canals and waiting to have oral surgery to remove two teeth. 9 months from my last injection. All my teeth hurt and are getting loose, I’m in pain and it hurts when I eat. My jaws are starting hurt and I’m scared. My health insurance doesn’t cover any of this so I’m looking at about 20k and just getting started. My Dentist office is closed because of the virus and I’m in the middle of a lot of bad fitting temporaries. I’m scared.

I was on Prolia for a couple of years and stopped taking it about a year ago after my doctor asked if I had been sick a lot since taking it! My answer was yes I had. I have since found out that both of my jaw joints are breaking down due to necrosis and will eventually need to be replaced. I am taking Baclofen to relax my jaw muscles so that I am able to open my jaw correctly. I have Crohns disease and Primary Immunoglobulin issues and should never have been prescribed this medication. While being on Prolia I had more incidents of the flu and Shingles. Just couldn’t stay well even though I do weekly at home immunoglobulin infusion treatments.

I am due for my second injection of Prolia today. After reading all these comments I am absolutely terrified of going ahead, Three months after having the first injection I was hospitalised for 6 days with severe diverticulitis. Prolia?… I don’t know. Is interesting how none the comments I am reading mention the situation we are facing in the world due to the Corona Virus. Is it because Prolia is a worse enemy ?….

Problem related to hip implant. I had a total hip replacement in 2011. Started receiving Prolia injections in 2016. Did very well until June 2017. Suddenly had onset hip pain, eventually diagnosed as peri- implant infection. Orthopedist said 4 oz. pus in area of joint, removed it, and put in temporary implant. I had to spend two months in a nursing home receiving I .v. Antibiotics. Then new hip implant about a month later. Followed by long recovery with months of PT. I truly believe Prolia was cause of all this.. Now on Fosamax. Not interested in lawsuit- just warning to others.

I was advised by my internist in 2017 that I was going into osteoporosis (after several meds were having no effect) and had my first shot(Prolia) in 2017 followed by two in 2018. I had no effect (except for teeth problems)from the first two but three days after the third shot, I experienced pain in the front of both legs (shins and thighs as well as around each kneecap). Three months later (2/19) I went back to the internist with my pain and he gave me a bone density test that showed anywhere from3%-10% improvement. But the pain continued to get stronger. By 5/19 my left hip was so painful that I could hardly walk/sleep. I was sent to a orthopedic team (back, hip,nerve) and found no reason for the pain in my legs or knees. My hip showed some deterioration but this did not bother me until the Prolia shot. I went thru six months of tests and no doctor would say that all my pain was caused by the shots. They did admit that the frontal pain from my waist down was not caused by the hip. Went back to the internist in 12/19 and he did every blood test to rule out every cause. All came back negative EXCEPT for the swollen muscle test that showed muscle inflammation, but couldn’t tell me where in my body. Because of all the pain, I would not take a fourth shot. There was some relief, both in my hip and thigh, from the three cortisones shots I have taken in the last six months but didn’t”t last long. I can’t walk more than a mile without significant pain. I feel I have lost over a year of my life and hope the longer this shot leaves my body, the pain will subside. My internist gave me gabapentin (300mg) in my Dec visit so I can sleep at night. Prolia should be given to those individuals as was intended – bone cancer or such severe osteoporosis that bone breaks are inevitable. This is not a preventative measure for the rest of us. I was never informed that this shot had to be continually given or, if stopped, the potential risk of fracture would increase. It also raised my blood pressure and cholesterol.

I had one injection of prolia in the Spring of 2019 and the second in the fall of 2019. After the first injection I had mild joint and muscle pain but that increased severely after the second. I have had X-rays, MRI, muscle relaxers, 15 steroid injections in the neck and back and nothing seems to help. I am so tired of this pain. I have always been a very healthy and active person and it seems like I have aged 20 years since taking Prolia! I am wondering if it will eventually go away since I will not take it again.

I have had 2 prolia shots starting in late 2018 ..my life is pretty well over. It started out, I lost all my energy. I can barely see out of my left eye.i keep a headache. My jaw bone hurts, its hard to eat.I have had an ulcer in my nose for a year and it’s getting worse and going to the other side.my immune system is gone. I have a thrush in my mouth and thru my whole system. It’s hard to eat.i don’t know what is going to happen next. I am 72 years old and my life was great until my Dr decided to give me Prolia. Now I don’t have a life. I really thought I was dying , and I may yet.

I had 2 prolia shots with in a year after the second shot I noticed my legs were hurting and my teeth and jaw were very painful, 3 mos. after the 2nd shot I developed C-diff a severe infection lost the ability to walk and move my arms . As the prolia wiped out my immune system white blood count off the charts heart racing and kidney problems spend 18 days in hospital and so much medicine drs took me off it I will never forget what that drug did to me still having problems no one should have to suffer like that I am only 69 was in good health. I was diagnosed with lung disease in Dec. 2019 where my initial symptons were shortness of breath and severe fatigue. I am now on oxygen and at 73 yrs of age feel hopeless. Started Prolia injection in Nov 2017 after a wrist fx. Hs of osteo-porosis. Now I am contemplating stem cell therapy which is not covered by my ins. After looking back at my health hs i am totally sure it is from the prolia injection. My drs do not know as Prolia does not list all of the side effects. I just know my body and believe Prolia is the cause of my illness.

I fx l wrist in nov 2017 and was put on Prolia 60mg every 6 months thru dec 2019. I noticed increased back and neck pain after walking but thought i was havingproblems due to lack of exercise after fx. In july 2019 started with shortness of breath and have been seeing a lung specialist since Oct 2019. r/o severe lung disease but the SOB IS THERE ALWAYS. Now recent loose tooth. even thou i have had problems worsen over time thought it was lack of exercise but it all started after taking Prolia. I told drs about symptons but they did not connect, even thou i never had this kind of pain before. I do have arthritis and fibromyralgia but everything hurts more now. i want to know why Prolia does not include these side effects as it was thought I had pulm fibrosis but recently r/o but lung infection?

My doctor kept insisting I have this injection even though I had no bone issues, I had one injection and began to get broken ribs! It’s been over three years and this continues (broken ribs) why is this not taken off the market?? It has changed my life and now I am worse for having even had one injection.

I have had 10 Prolia injections in the past 5 years. During this period, I had 11 dental implants, 6 before Prolia, and 5 after. All of the implants after starting Prolia failed. Each failure came with infection and jawbone damage.

Have been on Prolia since 2017 and just now finding out of all these side effects. I am so worried. My sister is having problems with uti’s pneumonoa and jaw problems. Iam scared, I will not take another shot. I have had Breast cancer and now i find out that this drug can cause cancer also.

P.s. from Alice. I also had a hip replacement that was also a side effect from Prolia. Can this drug be reversed now that I stopped taking it?…….my poor husband thought I was crazy crying all the time in pain, and he has to help me do my housework a lot of times. Thanks Prolia….
